MOLECULAR IDENTIFICATION OF SOME APHID SPECIES (HOMOPTERA; APHIDIDAE) BASED ON RFLP-PCR TECHNIQUE

This work includes, identification eight species of Aphids (Homoptera: Aphididae) which collected from the leaves different plants in many localities Erbil governorate Kurdistan region-Iraq period May till July 2022, these are: Chaitophorus salijaponicus, Aphis fabae, Macrosiphum rosae, Capitophorus carduinus, Myzus persicae, ruborum, punca, and gossypii. The mitochondrial cytochrome C Oxidase subunit I (COI) gene used for species. DNA was isolated, a band 550 bp mt COI amplified during PCR amplification. amplicons were digested with HinfI DdeI restriction enzymes. restricted fragments produced by RFLP technique proved agarose gel electrophoresis. results illustrated that given bands according to their cut sites. study presented studying aphids detect through RFLP-PCR using enzymes can distinguish some reliable results. REs could not all species, only discriminated persicae gossypii, but identified remain ruborum,within among other exactly. suggested provide full recognition profile
